HydroShear®
The best way to fragment DNA for library construction.
GeneMachines® HydroShear® offers the simplest, most reproducible, and most controllable method available for generating random DNA fragments. This software-driven device uses hydrodynamic shearing forces to fragment DNA strands.
With the HydroShear®, virtually any source of DNA at any concentration in volumes as small as 40μl can be randomly fractured to within a two-fold size distribution. For instance, if the desired target size is 2kb, the HydroShear® can be calibrated to produce a distribution centered on 2kb with 90% of the DNA falling between 1.3kb and 2.6kb – ideal for plasmid sequencing libraries. Furthermore, because of the reproducibility of the HydroShear® process, the distribution will be the same each day even with multiple users.

More about HydroShear®:
The HydroShear® uses hydrodynamic shearing forces to fragment DNA strands. The physical mechanism is fairly straightforward as shown in the image at the right.
- DNA in solution is passed through a tube with an abrupt contraction.
- As it approaches the contraction, the fluid accelerates to maintain the volumetric flow rate through the smaller area of the contraction.
- During this acceleration, drag forces stretch the DNA until it snaps. The DNA fragments until the pieces are too short for the shearing forces to break the chemical bonds. The flow rate of the fluid and the size of the contraction determine the final DNA fragment sizes.
System Components
The HydroShear® DNA shearing device includes the base unit, a 500 uL syringe, 4 standard shearing assemblies, wash solutions for 50 samples, the HydroShear® software, the HydroShear® User Manual© and Quick Start Guide©, tool kit, 1 power cord, 1 RS-232 cord, and connectors.
Sample Type Compatibility
The HydroShear® DNA shearing device effectively shears double-stranded DNA.
- Lengths Achievable Using the Standard Shearing Assembly: 1,000 bp to 9,000 bp
- Lengths Achievable with Custom Shearing Assemblies: Minimum: 650 bp, Maximum: 40,000 bp
- Maximum Concentration: Limited only by the maximum quantity of DNA that can be fully soluble in the desired sample volume. (Manufacturer validated for up to 0.25ug/uL)
- Volume: 40uL – 500 uL
Performance
- Run Time: Entire run including loading, running, and washing can be completed in 10 minutes.
- Repeatability: Shearing performance is independent of initial fragment length, sample volume, sample concentration, and user’s level of experience.
